Target description

Tumor necrosis factor receptor superfamily member 9 (UniProt: P20334; also known as 4-1BB ligand receptor, T-cell antigen 4-1BB, CD137) is encoded by the Tnfrsf9 (also known as Cd137, IIa, Ly63) gene (Gene ID: 21942) in murine species. CD137 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 24-187), a transmembrane domain (aa 187-208), and a cytoplasmic domain (aa 209-256). It is a member of the TNFR superfamily that serves as an activation-induced costimulatory molecule. Its expression is observed on activated T cells, NK cells, dendritic cells, eosinophils, mast cells, endothelial cells, and some tumor cells. It is rapidly expressed on activated CD8+ T cells and at lower levels on activated CD4+ T cells. Induction of CD137 expression on CD8+ T cells is reported to be highly sensitive to low levels of TCR stimulation. Engagement of CD137 on T cells stimulates their proliferation independently of CD28 and promotes IL-2 and IFN- secretion by Th effector cells and CTLs, respectively. Expression of CD137 is shown to negatively correlate with lymphocyte proliferation and positively with the degree of activation-induced cell death caused by mitogen overstimulation. Treg cells that infiltrate tumors are reported to express higher levels of CD137 and administration of clone LOB12.0 with agonistic function promotes T cell agonism to promote tumor rejection. Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
