General description

Laminin subunit alpha-5 (UniProt: O15230; also known as Laminin-10 subunit alpha, Laminin-11 subunit alpha, Laminin-15 subunit alpha) is encoded by the LAMA5 (also known as KIAA0533, KIAA1907) gene (Gene ID: 3911) in human. Laminins are a family of heterotrimeric extracellular glycoproteins consisting of alpha, beta, and gamma chains. Laminins mediate the attachment, migration and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. Thus far five alpha, three beta, and three gamma subunits are known, which assemble into at least 15 different laminin molecules. The alpha, beta, and gamma chains are bound to each other by disulfide bonds into a cross-shaped molecule comprising one long and three short arms with globules at each end. Alpha-5 is a subunit of laminin-10 (laminin-511), laminin-11 (laminin-521) and laminin-15 (laminin-523). It contains 22 laminin EGF-like domains, 5 5 laminin G-like domains, 1 laminin IV type A domain, and 1 laminin N-terminal domain. Laminin 5-alpha is expressed in heart, lung, kidney, muscle, pancreas, retina, and placenta. Very little or no expression is observed in brain and liver.

Specificity

Clone 4C7 reacts with the globular domain of the alpha5 chain of intact laminin and blocks the neurite stimulating activity of laminin.

Application

Anti-Laminin alpha5, clone 4C7 (Ascites Free), Cat. No. MAB1924-C, is a mouse monoclonal antibody that detects Laminin alpha 5 and has been tested for use in ELISA, Immunofluorescence, Immunohistochemistry, Immunoprecipitation, and neutralization.
Immunohistochemistry Analysis: A representative lot detected Laminin alpha 5 in immunostained basement membranes in O.T.C.-embedded fetal and adult human lung cryosections (Pierce, R.A., et al. (2000). Am. J. Respir. Cell Mol. Biol. 23(6):742-747).

Neutralizing Analysis: A representative lot detected Laminin alpha 5 in adhesion of sickle cell patients (Hb SS) red blood cells (RBCs) to human placental laminin-coated surface (Lee, S.P., et al. (1998). Blood. 92(8):2951-2958).

ELISA Analysis: A representative lot detected Laminin alpha 5 in recombinant human (rh) Laminin-10 ( 5 1 1) and Laminin-11 ( 5 2 1), but not Laminin-8 ( 4 1 1) (Wondimu, Z., et al. (2013). PLoS One. 8(1):e53648).

Immunofluorescence Analysis: A representative lot detected Laminin alpha 5 in immunoreactivity in human skin and kidney cryosections (Wondimu, Z., et al. (2013). PLoS One. 8(1):e53648).

Immunoprecipitation Analysis: A representative lot immunoprecipitated Laminin alpha 5 in Laminin-10 ( 5 1 1) from A549 culture medium (Wondimu, Z., et al. (2013). PLoS One. 8(1):e53648).
