General description

TGN46 is one of several proteins that utilize retrograde transport to travel between the trans-Golgi network (TGN) and the endocytic pathway. Retrograde transport involves the tethering, docking, fusion, and post translational modification of TGN proteins. Further studies are still required to elucidate the roles and mode of action for TGN46.

Specificity

This antibody recognizes the cytoplasmic domain of TGN46.

Immunogen

Epitope: Cytoplasmic domain
KLH-conjugated linear peptide corresponding to the cytoplasmic domain of human TGN46.

Application

Anti-TGN46 detects levels of TGN46 proteins & has been published & validated for use in wb, IP & ICC.

Western Blot Analysis: 2 µg/mL of this antibody detected TGN46 in 10 µg of NIH/3T3 cell lysate. Multiple uncharacterized bands may be observed in some cell lysates.

Immunocytochemistry Analysis: A 1:500 dilution from a representative lot detected TGN46 in A431 and HeLa cells.

Quality

Evaluated by Immunoprecipitation in HeLa cell lysate.

Immunoprecipitation Analysis: 0.2 µg of this antibody immunoprecipitated TGN46 in 500 µg of HeLa cell lysate (An uncharacterized band at ~29 kDa may be observed in some cell lysates.).

Target description

~110 kDa observed. This protein is observed at ~110 kDa, and it is subject to glycosylation (Khanobdee, K. et al. (2004). Mol Vis. 13(10):933-42.; Prescott, A. R., et al. (1997). Eur J Cell Biol. 72(3):238-246.).

The multispanning membrane protein vacuole membrane protein 1 (VMP1) marks and regulates endoplasmic reticulum (ER)-domains associated with diverse ER-organelle membrane contact sites. A proportion of these domains associate with endosomes during their maturation and remodeling. We found that these VMP1
