General description

Note: 4µg = 6000 units.
Native PKGIα purified from bovine lung. PKG is involved in regulation of smooth muscle relaxation, platelet function, and cell division. PKG is activated by binding of cGMP to regulatory subunits within the holoenzyme, but in contrast to PKA, the activation of PKG does not invlove physical dissociation of catalytic from the regulatory domains. The activated enzyme phosphorylates target proteins at serine and/or threonine residues that are found within the recognition motif, Arg-Lys-Arg-Ser*-Arg-Ala-Glu.

Unit Definition

One unit is defined as the amount of enzyme required to incorporote 1 pmol phosphate into the synthetic heptapeptide substrate, RKRSRAE, in 1 min at 30°C.

Physical form

In 150 mM NaCl, 25 mM β-mercaptoethanol, 10 mM K₂HPO₄, 1 mM EDTA, 12% sucrose, pH 6.8.

Reconstitution

Following initial thaw, aliquot and freeze (-70°C).
