General description

Netrin-1 is a secreted, bifunctional ligand belonging to the Netrin family; a group of proteins known to act as versatile, chemotropic axon guidance cues for CNS commissural and peripheral motor axons. Netrin-1 is necessary for spinal cord oligodendrocyte development. There are two receptors with which Netrin-1 is associated and that will cause it to be either a chemoattractant or a chemorepellant for axons. Receptor Deleted in Colorectal Cancer (DCC) regulates the attraction of axons to Netrin-1, and UNC5 mediates axon repulsion from Netrin-1. Studies have shown Netrin-1 to function as a survival factor through its relationship to its receptors, inhibiting the onset of apoptosis. Netrin-1 has also been implicated in colorectal tumorigenesis via its apoptotic regulatory role. Expression of Netrin-1 has been detected in adult axons and oligodendrocytes, and is also widely observed in developing embryonic mesodermal tissues and nervous system.

Quality

Evaluated by Western Blot in mouse brain tissue lysate.

Western Blot Analysis: 1:10,000 dilution of this antibody detected Netrin-1 on 10 µg of mouse brain tissue lysate.

~68 kDa observed. Characteristic proteolytic fragments of Netrin-1 found in the CNS run at lower molecular weights.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
