General description

4ME 16:0 Coenzyme A, also known as phytanoyl coenzyme A, is a coenzyme A derivative of phytanic acid. Phytanoyl coenzyme A is a metabolic intermediate of phytol degradation pathway. It is converted to 2-hydroxyphytanoyl-CoA by the enzyme phytanoyl-CoA 2-hydroxylase (PAHX). Accumulation of phytanic acid due to the deficiency of PAHX causes Refsum′s disease.

Phytanoyl-CoA 2-hydroxylase (PAHX), an iron(II) and 2-oxoglutarate-dependent oxygenase, catalyses an essential step in the mammalian metabolism of beta-methylated fatty acids. Phytanoyl-CoA was synthesised and used to develop in vitro assays for PAHX. The product of the reaction was confirmed as
Phytanoyl-coenzyme A hydroxylase deficiency -- the enzyme defect in Refsum's disease.
