Gene Information

human ... PROS1(5627)

General description

PROS1 (protein S alpha) gene is the active PS (Protein S) gene localized to human chromosome 3p 11.1 - 3q 1 1.2. Another gene PS β (PROS2) is a pseudogene. Exon I of this gene encodes the signal peptide, exon II encodes the propeptide and Gla domain, and exon III encodes a domain with a high aromatic amino acid content. The protein contains a thrombin-sensitive loop encoded by exon IV, four epidermal growth factor-like domains by exons V-VIII, and a sex hormone binding globulin-homologous domain encoded by the last seven exons.

Application

Anti-PROS1 antibody produced in rabbit has been used for global protein profiling to find new molecular biomarkers for common, multifactorial disorders.
Anti-PROS1 antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project . Each antibody is tested by immunohistochemistry against hundreds of normal and disease tissues. These images can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. The antibodies are also tested using immunofluorescence and western blotting. To view these protocols and other useful information about Prestige Antibodies and the HPA, visit sigma.com/prestige.

Biochem/physiol Actions

PROS1 (protein S alpha) gene encodes a vitamin K-dependent plasma glycoprotein that functions as a cofactor to activated protein C (APC) in the degradation of coagulation factors Va and VIIIa to inhibit blood coagulation in the presence of phospholipids and Ca2+ ions. The protein is active when it is circulating in the plasma in a free form. When bound to C4b-binding protein (C4b-BP), it circulates in the plasma in an inactive form. Only the active form serves as a cofactor to APC. Mutations in this gene may cause free protein S (type IIa) deficiency resulting in autosomal dominant hereditary thrombophilia.

Features and Benefits

Prestige Antibodies® are highly characterized and extensively validated antibodies with the added benefit of all available characterization data for each target being accessible via the Human Protein Atlas portal linked just below the product name at the top of this page. The uniqueness and low cross-reactivity of the Prestige Antibodies® to other proteins are due to a thorough selection of antigen regions, affinity purification, and stringent selection. Prestige antigen controls are available for every corresponding Prestige Antibody and can be found in the linkage section.

Every Prestige Antibody is tested in the following ways:
  • IHC tissue array of 44 normal human tissues and 20 of the most common cancer type tissues.
  • Protein array of 364 human recombinant protein fragments.
