Human Amelanotic Melanoma

Cell Line Description

This cell line has been derived by the selection of the human amelanotic melanoma cell line C32 for 6-thioguanine resistance. The parent cell line C32 was established from a melanoma from a 53 year old Caucasian male. The cells are resistant to 6-thioguanine and sensitive to HAT medium. They are deficient in hypoxanthine ? guanine phosphoribosyl transferase (HGPRT-). The cell line is also known as C32-r16TG. The cells are tumorigenic in nude mice inoculated subcutaneously with 10E7 cells.

Culture Medium

EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 0.050mM 6-thioguanine + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80 %) 1:2 to 1:4 i.e. seeding at 3-5x10,000 cells/cm2 using 0.25% trypsin/EDTA; 37 C; 5% CO2.
