Cell Line Description
HR5 cells were obtained after transfection of HeLa cells with plasmids coding for a reverse tetracycline-controlled transactivator encoding a nuclear localisation sequence (rt TA-nls) and transferring G418 resistance. This transactivator (a fusion between the Tet repressor and the activating domain of the herpes simplex virus VP16 transcription activation domain) can stimulate the expression of a reporter gene up to 10E5-fold. HR5 is the parent cell line for HR5-CL11 (Sigma Catalogue number.95091411).

Culture Medium
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) 1:5 to 1:20 i.e. seeding at 2x1,000 to 2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
