General description

Zinc finger protein Helios (Uniprot Q9UKS7; also known as Ikaros family zinc finger protein 2, Zinc finger DNA binding protein Helios, Zinc finger protein subfamily 1A member 2) is encoded by the IKZF2 (also known as ANF1A2, HELIOS, ZNF1A2, ZNFN1A2) gene (Gene ID 22807) in human. Helios, Ikaros, Aiolos, Eos, and Pegasus constitute a family of transcription factors characterized by six highly conserved C2H2 zinc fingers. Their four N-terminal zinc fingers mediate DNA binding, while the two C-terminal zinc fingers are responsible for homodimeric and heterodimeric protein interactions. Eos and Pegasus are expressed in a wide variety of tissues, whereas Helios, Ikaros, and Aiolos are limited to the hematopoietic system. Specifically, Helios expression is reported to be restricted to a subpopulation (approximately 70%) of Foxp3(+) T cells in mice and humans.

Specificity

Expected to react with isoform 1, 2 (Helios 1v), 3 (Helios del(Ex3,4)), 4 (Helios del(Ex6)v), 5 (Helios 1+2a,2b, Helios 1v+2a, Helios del(Ex3)+2a), 6 (Helios 1+5a, Helios 1+5a,5b), 7 (Helios del(Ex5)v), but not isoform 8 (Helios S).

Application

Flow Cytometry Analysis: 1.0 µg of this antibody from a representative lot detected Helios in 1X10E6 human Jurkat cells.
Flow Cytometry Analysis: A representative lot detected Helios immunoreactivity among the majority of CXCR5-positive cells within the CD4+Foxp3+ population of murine peripheral lymph node (LN) cells (Chung, Y., et al. (2011). Nat Med.17(8):983-988).
Flow Cytometry Analysis: A representative lot detected Helios immunoreactivity in the Foxp3-positive, but not Foxp3-negative, subpopulation of CD4+ cells from murine peripheral lymph node (LN) and human PBMCs (Thornton, A.M., et al. (2010). J Immunol.184(7):3433-3441).
