General description

The Estrogen Receptors (ER) belong to the nuclear receptor family that contains a ligand-binding domain that maintains the protein in an inactive conformation unless bound to the hormone. Two ERs are known, ERα, classically known as ER, and ERβ, described long after ERα. Activated Estrogen Receptor is a DNA-binding protein capable of inducing many changes in gene expression. The Estrogen Receptor is believed to play an important role in the growth and development of a subset of hormone dependent human breast cancers. ERβ is expressed in prostate, ovary, and other reproductive organs. Although its steroid specificity is very similar to that of the ERα, the molecular mechanisms regulating the transcriptional activity of ERβ may be distinct from those regulating ERα.

Specificity

Predicted to cross-react with rat, mouse, bovine and porcine based on sequence homology.
Recognizes Estrogen Receptor α. No cross-reactivity with Estrogen Receptor β.

Immunogen

Epitope: N-term
KLH-conjugated, synthetic peptide corresponding to amino acids 13-32 (C-ALLHQIQGNELEPLNRPQLK) of human Estrogen Receptor α. The immunizing sequence is identical in rat and mouse and has 19/20 identical amino acids in bovine and porcine.

Application

Anti-Estrogen Receptor α Antibody, clone 60C is an antibody against Estrogen Receptor α for use in WB & IP.
Immunoprecipitation:
4-10 μL of a previous lot of this clone immunoprecipitated Estrogen Receptor α from 1 mg of MCF-7 RIPA lysate.

Quality

Routinely evaluated by western blot analysis.

Western Blot Analysis:
This lot detected ERα at a 1:2,000 dilution in MCF-7 lysates via SDS-PAGE and transferred to PVDF (Immobilon-P).
