Specificity

Clone 1F4 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detect Sphingosine-1-phosphate transporter SPNS2. It targets an epitope within 23 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the internal region of human Sphingosine-1-phosphate transporter SPNS2.

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in Human heart tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected SPNS2 in Human heart tissue sections.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound SPNS2 peptide with a KD of 9.3 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SPNS2 in ACHN cells.

Target description

Sphingosine-1-phosphate transporter SPNS2 (UniProt: Q8IVW8; also known as Protein spinster homolog 2, SPNS2) is encoded by the SPNS2 gene (Gene ID: 124976) in human. SPNS2, a member of the large family of non-ATP-dependent organic ion transporter, is a multi-pass membrane protein that serves as a lipid transporter and specifically mediates the export of sphingosine-1-phosphate (S1P) and sphinganine-1-phosphate. It regulates levels of S1P and the S1P gradient that exists between the high circulating concentrations of S1P and low tissue levels that control lymphocyte trafficking. It is required for the egress of T-cells from lymph nodes during an immune response by mediating S1P secretion, which generates a gradient that enables activated T-cells to access lymph. It is also essential for the egress of immature B-cells from the bone marrow. SPNS2 also plays a role in innate immunity and SPNS2 knockout animals show reduced IFN- responses to infections due to the inability of natural killer cells to egress from lymph nodes. The deletion of SPNS2 is reported to impair humoral immune responses to immunization, reduced sensitivity to antigen-induced airway inflammation and hypersensitivity and attenuated delayed-type contact hypersensitivity. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
