Specificity

Clone 2F10 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects F4/80 (Adhesion G protein-coupled receptor E1). It targets an epitope within 15 amino acids from the N-terminal, extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the N-terminal region of human F4/80 (Adhesion G protein-coupled receptor E1).

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected F4/80 in Jurkat cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected F4/80 in Raji, NIH3T3, and human spleen tissue lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected F4/80 in Jurkat cells.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected F4/80 in one million Jurkat cells.

Affinity Binding Assay: A representative lot of this antibody bound F4/80 peptide with a KD of 2.0 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: 1:100 from a representative lot detected F4/80 in human spleen and human bone marrow tissue sections.

Anti-F4/80, clone 2F10 ZooMAb®, Cat. No. ZRB1277, is a recombinant Rabbit monoclonal antibody that targets F4/80 and is tested for use in Affinity Binding Assay, Flow Cytometry, Immunocytochemistry, Immunohistochemistry, and Western Blotting.

Target description

Adhesion G protein-coupled receptor E1 (UniProt: Q14246; also known as EGF-like module receptor 1, EGF-like module-containing mucin-like hormone receptor-like 1, EMR1 hormone receptor, F4/80) is encoded by the ADGRE1 (also known as EMR1, TM7LN3) gene (Gene ID: 2015) in human. F4/80 is multi-pass membrane glycoprotein of the G-protein coupled receptor 2 family that contains four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. It also has six EGF-like domains in the N-terminal region followed by a serine/threonine-rich stalk region. F4/80 is involved in cell adhesion and in cell-cell interactions involving the immune system. F4/80 is reported to be a pan-macrophage marker mouse to detect the distribution and localization of tissue macrophages and to examine the phenotypic heterogeneity and activation state of resident macrophages. Although not required for normal macrophage development in murine species, F4/80 is shown to be essential for the induction of efferent CD8+ regulatory T-cells, which is needed for peripheral tolerance. In human F4/80 expression is restricted to eosinophils. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
