General description

Phosphatidic acid (PA) is a simple anionic cellular glycerophospholipid possessing a phosphomonoester polar head group and two long hydrophobic acyl chains. 7-nitro-2-1,3-benzoxadiazol-4-yl (NBD) PA is a fluorophore labeled phospholipid having 7-nitrobenz-2-oxa-1,3-diazol-4-yl (NBD) at the sn-2 fatty acid of PA.

Application

16:0-12:0 NBD PA or 1-palmitoyl-2-{12-[(7-nitro-2-1,3-benzoxadiazol-4-yl)amino]dodecanoyl}-sn-glycero-3-phosphate (ammonium salt) is suitable:
  • to incubate cells for the imaging of lyso phosphatidylcholine (PC) in P. falciparum live cells
  • as a component of separation buffer for the separation of phosphoinositides and sphingolipids in the microchip devices
  • in cytidine diphosphate diacylglycerol (CDP-DAG) synthase activity assay

Biochem/physiol Actions

Phosphatidic acid (PA) influences membrane fusion and membrane fission events and membrane curvature. It acts as a signaling molecule for various cellular processes including membrane trafficking and sperm hyper-activation.

CDP-diacylglycerol (CDP-DAG) is central to the phospholipid biosynthesis pathways in cells. A prevailing view is that only one CDP-DAG synthase named Cds1 is present in both the endoplasmic reticulum (ER) and mitochondrial inner membrane (IM) and mediates generation of CDP-DAG

Phosphatidic acid (PA) is the simplest cellular glycerophospholipid characterized by unique biophysical properties: a small headgroup; negative charge; and a phosphomonoester group. Upon interaction with lysine or arginine, PA charge increases from -1 to -2 and this change stabilizes protein-lipid
