Specificity

Clone 1F9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects CD41/Integrin 2b. It targets an epitope within 21 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the extracellular domain of human CD41/Integrin 2b.

Application

Quality Control Testing

Evaluated by Western Blotting in Human spleen tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD41/ITGA2B in Human spleen tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD41/ITGA2B in Human liver tissue lysate.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected CD41/ITGA2B in one million Human peripheral blood mononuclear cells (PBMC).

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD41/ITGA2B in A549 cells.

Affinity Binding Assay: A representative lot of this antibody bound CD41/ITGA2B peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CD41/ITGA2B in Human bone marrow tissue sections.

Target description

Integrin alpha-IIb (UniProt: P08514; also known as GPalpha IIb, GPIIb, Platelet membrane glycoprotein IIb, CD41) is encoded by the ITGA2B (also known as GP2B, ITGAB) gene (Gene ID: 3674) in human. CD41/ITGA2B is a single-pass type I membrane glycoprotein of the integrin chain family. It is synthesized with a signal peptide (aa 1-31), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 32-993), a transmembrane domain (aa 994-1019), and a short cytoplasmic domain (aa 1020-1039). Along with β3 it serves as a receptor of fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. It recognizes the sequence R-G-D in a wide array of ligands. Following activation, integrin IIb/β3 brings about platelet/platelet interaction through binding of soluble fibrinogen. This step leads to rapid platelet aggregation, which physically plugs ruptured endothelial cell surface. Mutations in ITGA2B gene have been linked to Glanzmann thrombasthenia 1 (GT1), a disorder characterized by failure of platelet aggregation, absent or diminished clot retraction, and mucocutaneous bleeding of mild-to-moderate severity. Three isoforms of CD41/ITGA2B have been described that are produced by alternative splicing. Isoform 1 and 2 have been identified in platelets and megakaryocytes, but not in reticulocytes or in Jurkat and U-937 cell lines. Isoform 3 is expressed by leukemia, prostate adenocarcinoma, and melanoma cells but not by platelets or normal prostate or breast epithelial cells. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
