Specificity

Clone 1C7 is a ZooMAb® rabbit recombinant monoclonal antibody that detects FIS1. It targets an epitope within 24 amino acids from the N-terminal cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal, cytoplasmic domain of human FIS1.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting: A 1:10,000 dilution of this antibody detected FIS1 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: 1:1,000-10,000 dilutions from a representative lot detected FIS1 in lysates from Raji cells and mouse liver tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected FIS1 in human kidney tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected FIS1 in HeLa cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Mitochondrial fission 1 protein (UniProt: Q9Y3D6; also known as FIS1 homolog (hFis1), Tetratricopeptide repeat protein 11 (TPR repeat protein 11)) is encoded by the FIS1 (also known as TTC11, CGI-135) gene (Gene ID: 51024) in human. FIS1 is a small, integral mitochondrial outer membrane protein characterized by a tetratricopeptide repeat (TPR) domain which facilitates protein-protein interactions. The cytosolic domain interacts with other proteins involved in mitochondrial fission. It acts by recruiting cytosolic factors such as dynamin-related protein 1 (DRP1), facilitating the assembly of the fission machinery that constricts and divides mitochondria. This process is crucial for mitochondrial distribution and health, affecting cellular energy production and apoptosis. Dysregulation of FIS1 expression or activity is associated with neurodegenerative diseases such as Alzheimer′s and Parkinson′s, where impaired mitochondrial dynamics contribute to cellular dysfunction and death. Additionally, FIS1 has been linked to metabolic disorders, as proper mitochondrial function is crucial for energy balance and metabolism. In cancer, altered mitochondrial fission can affect apoptosis and cellular proliferation, implicating FIS1 in tumorigenesis and cancer progression. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
