Specificity

Clone 2G18 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Potassium voltage-gated channel subfamily KQT member 2 (KCNQ2). It targets an epitope within 20 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal, cytoplasmic domain of human Potassium voltage-gated channel subfamily KQT member 2 (KCNQ2).

Application

Quality Control Testing

Evaluated by Western Blotting in Mouse brain microsomal preparation.

Western Blotting Analysis: 1:10,000 of this antibody detected KCNQ2 in Mouse brain microsomal preparation.

Tested applications


Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected KCNQ2 in human brain tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound KCNQ2 with a KD of 1.0 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected KCNQ2 in E18 rat cortical cells.

Anti-KCNQ2, clone 2G18 ZooMAb®, Cat. No. ZRB1299, is recombinant Rabbit monoclonal antibody that detects KCNQ2 and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry, and Western Blotting.

Target description

Potassium voltage-gated channel subfamily KQT member 2 (UniProt: O43526; also known as KQT-like 2, Neuroblastoma-specific potassium channel subunit alpha KvLQT2, Voltage-gated potassium channel subunit Kv7.2, KCNQ2) is encoded by the KCNQ2 gene (Gene ID: 3785) in human. KCNQ2 is a multi-pass membrane protein with four cytoplasmic domains, six transmembrane domains, and four extracellular domains. It is highly expressed in areas containing neuronal cell bodies and low expression is observed in spinal cord and corpus callosum. KCNQ2 associates with KNNQ3 to form a potassium channel with essentially identical properties to the channel underlying the native M-current, a slowly activating and deactivating potassium conductance, which plays a role in determining the subthreshold electrical excitability of neurons and the responsiveness to synaptic inputs. It is also important in regulation of neuronal excitability. Mutations in KCNQ2 gene are usually inherited in an autosomal-dominant manner in epileptic syndromes and KCNQ2-associated seizures are observed as early as the first week after birth. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
