Specificity

Clone 1G17 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Period circadian protein homolog 2 (PER2). It targets an epitope within 21 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal region of human Period circadian protein homolog 2 (PER2).

Application

Anti-PER2, clone 1G17 ZooMAb, Cat. No. ZRB1076, is a recombinant Rabbit monoclonal antibody that specifically targets PER2 and has been tested for use in Affinity Binding Assay, Immunocytochemistry, and Immunohistochemistry (Paraffin).
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PER2 in human retina tissue sections.

Immunocytochemistry Analysis: A 1:1000 dilution from a representative lot detected PER2 in HeLa cells.

Affinity Binding Assay: A representative lot of this antibody bound PER2 peptide with a KD of 7.2 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Period circadian protein homolog 2 (UniProt: O15055; also known as hPER2, Circadian clock protein PERIOD 2, PER2) is encoded by the PER2 (also known as KIAA0347) gene (Gene ID: 8864) in human. PER2 is a homodimeric protein that is widely expressed and is high levels are found in skeletal muscle and pancreas. It serves as a transcriptional repressor and is a core component of the circadian clock along with other clock components (CLOCK, NPAS2, ARNTL/BMAL1, ARNTL2/BMAL2, PER1, PER2, PER3, CRY1 and CRY2) it plays a critical role in rhythm generation. Its levels oscillate diurnally, and its rhythmic levels are critical for the generation of circadian rhythms in central as well as peripheral clocks. Transcription factors, CLOCK or NPAS2 and ARNTL/BMAL1 or ARNTL2/BMAL2, form the positive limb of the feedback loop, act in the form of a heterodimer and activate the transcription of core clock genes and clock-controlled genes. The core clock genes: PER1/2/3 and CRY1/2 which are transcriptional repressors form the negative limb of the feedback loop and interact with the CLOCK/NPAS2, ARNTL/BMAL1, ARNTL2/BMAL2 heterodimer inhibiting its activity. PER2 shuttles between the nucleus and cytoplasm and this shuttling is affected by interaction with other circadian core oscillator proteins and/or by phosphorylation. It translocates to the nucleus after phosphorylation by CSNK1D or CSNK1E. PER2 contains a nuclear localization signal (aa 789-805) and three nuclear export signals (aa 111-120, 462-471, and 989-996). Mutations in PER2 gene have been linked to advanced sleep phase syndrome. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
