Cell Line Origin

Human megakaryoblastic leukaemia

Cell Line Description

Derived from the bone marrow of a 55 year old male with chronic myelogenous leukaemia. All karyotypes show the Philadelphia chromosome. Platelet GP IIb/IIIa antigen is expressed on more than 92% of cells. Cytoplasmic expression of GP Ib antigen was not detected on the surface of cells but was present in the cytoplasm 20% of cells. BA-1 antigen is expressed in almost all cells. FVIII-related antigen was not detected on the surface cells but was present in cytoplasm.

Application

Studies of physiological mechanisms governing the differentiation of human megakaryocyte and platelet release; the study of the biochemistry (and differentiation) of erythroid and myeloid cells.

Culture Medium

RPMI 1640 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Subcultures are prepared by diluting the cell suspension into fresh medium at a ratio of 1:2 to 1:3. Cells should be seeded at 2-4 x 105 cells/ml. Doubling time is 36-48 hours. 5% CO2; 37°C.
