General description

Clathrin, light chain A also known as LCA is a soluble protein composed of heavy and light chain. Clathrin is a protein that plays a major role in the formation of coated vesicles. It coats vesicles from brain, adrenal medulla and non-secreting lymphoma cell line. Clathrin-coated vesicles (CCVs) are major carriers for endocytic cargo and maintain important intracellular trafficking events.

Immunogen

synthetic peptide (amino acids 23-44 in bovine) common to the N-terminus of mammalian clathrin light chains, LCa and LCb, conjugated to KLH. This region is not involved in heavy chain binding and is completely conserved in all mammals.

Application

Monoclonal Anti-Clathrin Light Chain antibody produced in mouse is suitable for the following applications:
  • Immunocytochemistry
  • Immunoprecipitation
  • Microarrary
  • Western blotting at a concentration of 1-2μg/mL using whole rat brain homogenate

Biochem/physiol Actions

Clathrin requires cofactors like ADP-ribosylation factor 6 (ARF6) and adaptin for regulation of receptors. Clathrin light chains (CLCs) are not required for clathrin-mediated endocytosis but plays a critical role for clathrin-mediated trafficking between the TGN and the endosomal system whereas, clathrin heavy chain provides the structural backbone of the clathrin coat. CLCs in mammalian cells function in intracellular membrane trafficking by acting as recruitment proteins for huntingtin-interacting protein 1-related (HIP1R), enabling it to regulate actin assembly on clathrin-coated structures.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
