General description

C-C chemokine receptor type 2 (UniProt P41597; also known as C-C CKR-2, CC-CKR-2, CCR-2, CCR2, CD192, MCP-1-R, Monocyte chemoattractant protein 1 receptor) is encoded by the CCR2 (also known as CMKBR2) gene (Gene ID 729230) in human. CC chemokine receptors (CCRs) are 7-transmembrane G protein-coupled receptors (GPCRs) that bind CC chemokines (CCLs) and mediate their biological activities. CCRs and CCLs play a central role in physiological and pathological recruitment of immune cells. CCR2 can interact with CCL2, CCL8 and CCL16, it is found on the surface of monocytes, activated memory T cells, B cells, and basophils in humans, and also in peritoneal macrophages in mice. CCR2 plays an important role in the recruitment of monocytes/macrophages and T cells. CCR2-knockout mice show a partial defect in macrophage function, decreased Th1 immune response, reduced delayed-type hypersensitivity reaction, and are more susceptible to pathogenic bacteria infections. Human CCR2 is a 7-transmembrane (a.a. 43-70, 81-100, 115-136, 154-178, 207-226, 244-268, 286-309) GPCR with an N-terminal extracellular domain (a.a. 1-42) and a C-terminoal cytoplasmic domain (a.a. 310-374).

Specificity

Clone 8C3.1 targets the N-terminal extracelluar domain of human CCR2 spliced isoforms 1&2 reported by Uniprot (P41597).

Immunogen

GST-tagged recombinant human N-terminal extracellular domain.

Application

Flow Cytometry Analysis: 2 µL from a representative lot detected surface CCR2 expression among the gated lymphocytes population from one million human PMBCs.

This mouse monoclonal Anti-CCR2 Antibody, clone 8C3.1, Cat. No. MABF299 is validated for use in Flow Cytometry and Western Blotting for the detection of CCR2.

Quality

Evaluated by Western Blotting in U937 cell lysate.

Western Blotting Analysis (WB): A 100,000 dilution of this antibody detected CCR2 in 10 µg of human lymphoma U937 monocyte lysate.

Target description

~40-50 kDa observed. 41.92 kDa (isoform 1) and 41.06 kDa (isoform 2) calculated. Larger-than-calculated sizes and smeary banding pattern are consistent with the detection of differentially glycosylated CCR2 species. Uncharacterized bands may be observed in some lysate(s).
