General description

Contactin-2 (UniProt P22063; also known as Axonal glycoprotein TAG-1, Axonin-1, Transient axonal glycoprotein 1, TAX-1) is encoded by the Cntn2 (also known as Tax1) gene (Gene ID 25356) in rat species. Proper localization of the voltage-gated Na+ channels to the node of Ranvier and voltage-gated K+ channels to the juxtaparanode (JXP) along myelinated axons is essential for the fast propagation of action potentials. Localization of clustered ion channels in specific membrane domains depends on specific cell adhesion molecules (CAMs), as well as on the integrity of neighboring domains. Contactin-2/TAG-1 and Caspr2 form a membrane scaffold complex that functions to cluster Kv1 potassium channels at the JXP. Deletion of either of these CAMs causes Kv1 channels to disappear from the JXP and to redistribute along the internode. Contactin-2/TAG-1 is initially produced with an N-terminal signal peptide (a.a. 1-30) and a C-terminal propeptide (a.a. 1016-1040) sequence, the removal of which yields the mature GPI-anchored membrane glycoprotein.

Epitope: Fibronectin type-III domains 1-4.
Fc-tagged recombinant protein corresponding to the Fibronectin type-III domains 1-4 of rat TAG-1.

Application

Anti-TAG-1 Antibody is an antibody against TAG-1 for use in Western Blotting, Immunocytochemistry.
Immunocytochemistry Analysis: Representative lots detected TAG-1 immunoreactivity at the juxtaparanodal (JXP) region of teased sciatic nerves from wild-type, but not Caspr−/−, Caspr2−/−, or Caspr−/−Caspr2−/− double knockout, mice. Reexpressing full-length Caspr2, but not 4.1-binding sequence (d4.1) deletion Caspr2 mutant, in Caspr2−/− transgenic mice restored JXP TAG-1 immunoreactivity (Gordon, A., et al. (2014). J. Neurosci. 34(45):14820 –14826; Horresh, I., et al. (2010). J. Neurosci. 30(7):2480–2489).
Immunocytochemistry Analysis: A representative lot detected a loss of TAG-1 immunoreactivity at the juxtaparanodal (JXP) region of teased sciatic nerves isolated from 4.1B−/− mice lacking the cytoskeletal adapter protein 4.1B (Horresh, I., et al. (2010). J. Neurosci. 30(7):2480–2489).

Western Blotting Analysis: A 1:5,000 dilution of this antibody detected TAG-1 in 10 µg of mouse brain tissue lysate.

~140 kDa observed. Target band appears larger than the calculated molecular weight of 107.3 kDa due to glycosylation.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
