General description

Mis12 complex is composed of four subunits, Protein MIS12 homolog, Polyamine-modulated factor 1, Kinetochore-associated protein DSN1 homolog, and Kinetochore-associated protein NSL1 homolog (UniProt: Q9H081, Q6P1K2, Q9H410, Q96IY1, respectively) that are encoded by genes known as MIS12 (Gene ID: 79003), PMF1 (Gene ID: 100527963), DSN1 (Gene ID: 79980), and NSL1 (also known as C1orf48, DC31, DC8, MIS14) (Gene ID: 25936) in human. The MIS12 complex is a protein interaction hub for outer kinetochore assembly. This complex acts as the primary microtubule-binding interface at kinetochores and provides a platform to recruit regulatory proteins. In human Mis12 complex subunits are shown to localize with centromere protein A (CENP-A) at inner kinetochores and internally to Ndc80 at outer kinetochores. Mis12 complex plays an essential role in chromosome segregation in vertebrates and contributes to mitotic kinetochore assembly. Reduced levels of Mis12 complex proteins are shown to result in chromosome alignment defects in both human and chicken cells, but spindle bipolarity is not disturbed. (Ref.: Kline SL et al (2006). J. Specificity

This polyclonal antibody detects Mis12 complex in human. It will also detect individual subunits (Mis12, PMF1, Dsn1, Nsl1) in Western Blotting.

Application

Anti-Mis12 Complex Antibody, Cat. No. ABE2585, is a highly specific rabbit polyclonal antibody that targets Mis12 complex and has been tested in Immunocytochemistry, Immunofluorescence, and Western Blotting.
Immunocytochemistry Analysis: A 1:1,100 dilution from a representative lot detected Mis12 complex in HeLa cells.

Immunofluorescence Analysis: A representative lot detected Mis12 complex in HeLa cells (Rago, F., et. al. (2015). Curr Biol. 25(5):671-7).

Western Blotting Analysis: 2.2 µg/mL of this antibody detected Mis12 complex in 1 µL of recombinant Mis12 complex.

24.10, 23.34, 40.07, and 32.16 kDa calulated for Mis12, PMF1, DSN1, and NSL1, respectively. Uncharacterized bands may be observed in some lysate(s).

Storage and Stability

Stable for 1 year at -20°C from date of receipt.

Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
