General description

Menin (UniProt O00255) is a nuclear protein encoded by the MEN1 gene (also known as MEAI, SCG2) (Gene ID 4221) in human. MEN1 is a tumor suppressor gene frequently mutated in the familial multiple endocrine neoplasia type I (MEN-1) cancer syndrome characterized by tumors in multiple endocrine organs including the parathyroid, pancreatic islets, and the pituitary. Menin mediates cell proliferation and differentiation actions of transforming growth factor-beta (TGF-beta) ligand family members. Menin is reported to associate with histone methyltransferases (HMT) MLL1 (KMT2A) and MLL4 (KMT2B) to form menin-HMT complexes in both human and murine model systems. In addition, menin is also shown to be involved in DNA repair with FANCD2.

Specificity

Expected to react with all three spliced variants: Isoform 1 (Long), Isoform 2 (Short), and Isoform 3.

KLH-conjugated linear peptide corresponding to the C-terminus of human Menin/MEN1.

This Anti-Menin/MEN1 Antibody is validated for use in Western Blotting for the detection of Menin/MEN1 .
Western Blotting Analysis: A representative lot detected endogenous Menin in TGP61 nuclear extract, as well as exogenously expressed Menin in transfected NIH3T3 cells (Jin, S., et al. (2003). Cancer Res. 63(14):4204-4210).

Quality

Evaluated by Western Blotting in Tamoxifen treated MEF 545T and MEF 545D cell lysate.

Western Blotting Analysis: A 1:20,000 dilution of this antibody detected Menin/MEN1 in 10 µg of Tamoxifen treated MEF 545T and MEF 545D cell lysate.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
