General description

Specificity

Clone 1I9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Transcription factor SOX-5. It targets an epitope within 24 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal region of human Transcription factor SOX-5.

Application

Quality Control Testing

Evaluated by Western Blotting with DDK-tagged, full-length, Human SOX5.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected DDK-tagged, full-length, recombinant Human SOX5.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SOX5 in MCF7 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SOX5 in Human prostate tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound SOX5 with a KD of 4.5 x 10-7 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Transcription factor SOX-5 (UniProt: P35711; also known as SOX5) is encoded by the SOX5 gene (Gene ID: 6660) in human. SOX (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. SOX5 is a transcription factor that is involved in in chondrocytes differentiation and cartilage formation. It is reported to specifically bind the 5′-AACAAT-3′ DNA motif present in enhancers and super-enhancers and promotes expression of genes important for chondrogenesis, including cartilage matrix protein-coding genes, such as COL2A1 and AGC1. It is required for overt chondrogenesis when condensed pre-chondrocytes differentiate into early-stage chondrocytes. Together with SOX6, it maintains a pool of highly proliferating chondroblasts between epiphyses and metaphyses, to form columnar chondroblasts and delay chondrocyte pre-hypertrophy. SOX-5 has also been linked to the migration and invasion of various cancer cells, including hepatocellular carcinoma, melanoma, and prostate cancer. It is also shown to promote the migration and invasion of fibroblast-like synoviocytes in part by regulating MMP-9 expression in collagen-induced arthritis. Mutations in SOX5 gene have been linked to Lamb-Shaffer syndrome, an autosomal dominant, neurodevelopmental disorder that is characterized by developmental delay, intellectual disability, and motor impairment. Five isoforms of SOX5 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
