Specificity

Clone 1N23 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects cIAP2/BIRC3. It targets an epitope within 23 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the N-terminal half of human Baculoviral IAP repeat-containing protein 3 (cIAP2/BIRC3).

Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected cIAP2/BIRC3 in Raji cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected cIAP2/BIRC3 in Ramos cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound cIAP2/BIRC3 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected cIAP2/BIRC3 in Ramos cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected cIAP2/BIRC3 in human testis tissue sections.

Target description

Baculoviral IAP repeat-containing protein 3 (UniProt: Q13489; also known as EC:2.3.2.27, Apoptosis inhibitor 2, API2, Cellular inhibitor of apoptosis 2, C-IAP2, IAP homolog C, Inhibitor of apoptosis protein 1, hIAP-1; hIAP1, RING finger protein 49, RING-type E3 ubiquitin transferase BIRC3, TNFR2-TRAF-signaling complex protein 1) is encoded by the BIRC3 (also known as API2, MIHC, RNF49) gene (Gene ID: 330) in human. cIAP2/BIRC3 is a multi-functional zinc-binding protein that regulates the activity of caspases and apoptosis and modulates inflammatory signaling and immunity, cell proliferation, invasion, and metastasis. It acts as an E3 ubiquitin-protein ligase and regulates NF-kB signaling by acting as a positive regulator of the canonical pathway and suppresses constitutive activation of non-canonical NF-kB signaling. Via its E3 ubiquitin-protein ligase activity it can target RIP kinase 1, 2, 3, and 4, caspase 3, 7, 8,  and TRAF1, and BCL10. cIAP2/BIRC3 contains three baculoviral IAP repeat (BIR) domains (aa 29-96; 169-235; 255-322) that are involved in protein-protein interactions. Each BIR domain contains of 66-67 amino acids, whose fold is stabilized by a zinc atom that is coordinated by one histidine and three cysteine residues. It also has a caspase recruitment (CARD) domain (aa 439-529) and a C-terminal Ring zinc-finger domain (aa 557-592). Its high expression is observed in fetal lung and kidney. In the adult, its expression is mainly observed in lymphoid tissues. cIAP2/BIRC3 plays a role as a tumor suppressor and its deficiency is associated with poor prognosis and resistance to chemotherapy.  Mutations in BIRC3 gene or its deletion is reported in some cases of chronic lymphocytic leukemia (CLL). This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
