Specificity
Clone 1N22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Rab8A. It targets an epitope within 17 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human Ras-related protein Rab-8A (Rab8A).
Application
Quality Control Testing
Evaluated by Western Blotting in MCF-7 cell lysate.
Western Blotting Analysis: A 1:3,000 dilution of this antibody detected Rab8A in MCF-7 cell lysate.
Tested applications
Western Blotting Analysis: A 1:3,000 dilution from a representative lot detected Rab8A in lysates from PC12 and HCT116 cells and from Human kidney tissue lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Rab8A in human kidney tissue sections.
Immunocytochemistry Analysis: A 1:300 dilution from a representative lot detected Rab8A in MCF-7 cells.

Target description
Ras-related protein Rab-8A (UniProt: P61006; also known as Oncogene c-mel) is encoded by the RAB8A (also known as MEL, RAB8) gene (Gene ID: 4218) in human. The small GTP-binding protein Rab8 is involved in basolateral transport in epithelial cells and in dendritic transport in neurons. It is also involved in trafficking of neurotransmitter receptors in postsynaptic terminal and the formation of outer photoreceptor segments in the retina. Rab8A is a ubiquitous, lipid-anchor protein that functions in multiple recycling pathways through various effectors. It is synthesized with a propeptide (aa 205-207) that is subsequently removed to generate the mature form. In its GDP-bound form, it is present in the perinuclear region and in GTP-bound form it displays polarized distribution to distal regions of cell protrusions. Together with MYO5B and RAB11A, it participates in epithelial cell polarization. It undergoes phosphorylation at threonine 72 by leucine-rich repeat kinase 2 (LRRK2) in its switch II helix, which prevents the association of Rab regulatory proteins, including CHM, CHML, and Rab GDP dissociation inhibitors GDI1 and GDI2. Non-phosphorylated form of Rab8A is shown to be predominantly localized in the cytoplasm whereas phosphorylated Rab8A localizes to the membrane. Rab8a is also reported to recruit PI3K and together they function to regulate Akt signal transduction generated by toll like receptor 4 (TLR4). It binds to two residues in the receptor binding domain (RBD) of PI3K that are reported to be critical and necessary for Ras binding to PI3K . Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
