General description
Histone H3.1t (UniProt: Q16695; also known as H3/t, H3t, H3/g) is encoded by the HIST3H3 (also known as H3FT) gene (Gene ID: 8290) in human. Histone H3 has two main variants, H3.1 and H3.3, which show different genomic localization patterns in animals. The H3.1 and H3.3 complexes also possess distinct histone chaperones, CAF-1 and HIRA, which play important role in mediating DNA-synthesis-dependent and -independent nucleosome assembly. Histone H 3.1 is a core component of nucleosome that is present only in mammals. It is expressed during S phase, then its expression decreases significantly as cell division slows down during the process of differentiation. Histone H 3.1 expression is shown to be replication dependent. It serves as the canonical histone, which is incorporated during DNA replication. Histones are dynamic proteins that undergo multiple types of post-translational modifications, including acetylation, phosphorylation, methylation, and ubiquitination. Histone 3.1 is also shown to undergo serotonylation of glutamine at position 5 (Q5ser) in organisms that produce serotonin. It has been reported that Histone H3 can also undergo dopaminylation ((H3Q5dop), which can potentiate the binding of adjacent histone post-translational modification interacting proteins and play a vital role in dopaminergic neuronal transcription. Anti-H3Q5dopaminyl, Cat. No. ABE2588, is a rabbit polyclonal antibody that detects Histone H3 dopaminylated on glutamine 5 and is tested for use in Dot Blot, Peptide Inhibition Assay, and Western Blotting.

Western Blotting Analysis: A 1:500 dilution of this antibody detected recombinant Histone H3 dopaminylated on glutamine 5 with 2.5 mM dopamine, but not with one treated with transglutaminase 2 (0.5 mg).
