General description

Transforming acidic coiled-coil-containing protein 3 (UniProt Q9Y6A5; also known as ERIC-1 and AINT) is encoded by the TACC3 (also known as ERIC1) gene (Gene ID 10460) in human. Human TACC3 gene localizes close to the FGFR3 gene in 4p16, a region that is disrupted in multiple myeloma. TACC proteins have emerged as important players in the complex process of regulating microtubule dynamics during cell division. Three family members have been identified in humans: TACC1, TACC2, and TACC3. TACC3 plays a role in the microtubule-dependent coupling of the nucleus and the centrosome and is involved in the processes that regulate centrosome-mediated interkinetic nuclear migration of neural progenitors. TACC3 is expressed in relatively few adult tissues, but it shows elevated levels in testis and ovary, and in the hematopoietic lineages. During mouse development, TACC3 is present in all the embryonic stages and particularly in proliferating tissues. A comparison of the human and mouse TACC3 proteins shows a sequence conservation within the first 103 amino acids at the N-terminal region and the C-terminal 318 amino acids, which includes the coiled coil domain. Specificity

This polyclonal antibody specifically detected the TACC3 target band in untransfected, but not TACC3 shRNA-transfected, human cells (Rao, R., et al. (2014). Oncogene. 33(33):4242-4252).

Immunogen

Synthetic peptide corresponding to an internal sequence within the N-terminal half of human TACC3 (Rao, R., et al. (2014). Oncogene. 33(33):4242-4252).

Application

Anti-TACC3, Cat. No. ABE1942, is a highly specific rabbit polyclonal antibody that targets TACC3 and has been tested in Immunocytochemistry and Western Blotting.
Immunocytochemistry Analysis: A 1:650 dilution from a representative lot detected TACC3 in 4% paraformaldehyde-fixed, 0.3% Triton X-100-permeabilized HeLa cells.

Immunocytochemistry Analysis: A representative lot detected TACC3 spindle immunoreactivity in untransfected, but not TACC3 siRNA-transfected, SKOV-3 and OVCAR3 ovarian cancer cells (Rao, R., et al. (2014). Oncogene. 33(33):4242-4252).

Western Blotting Analysis: A representative lot detected much higher TACC3 expression in SKOV-3, OVCAR3 ovarian cancer cells than in non-cancerous hTERT-RPE, MCF10A, and 184B5 cells. Cellular TACC3 siRNA delivery abolished target band detection (Rao, R., et al. (2014). Oncogene. 33(33):4242-4252).

Quality

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: 1 µg/mL of this antibody detected TACC3 in 10 µg of HEK293 cell lysate.

~130 kDa observed. 90.23 kDa (Met1 removed) calculated. Uncharacterized bands may be observed in some lysate(s).

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
