For immunocytochemistry, HeLa cells were fixed with 95% ethanol/5% acetic acid and treated with 5 mM sodium butyrate, an inhibitor of histone deacetylases, for 24 h. 10-500 ng/ml of antibody detected acetylated histone H3 in acid-extracted proteins from HeLa cells. Using sodium butyrate, an inhibitor of histone deacetylases, enhances detection of acetylated histone H3. Higher concentrations of antibody also detected unacetylated histone H3. Preliminary evidence from an independent library suggests that phosphorylation of Ser10 on histone H3 may disrupt epitope recognition by this antibody. This antibody has also been reported to work for immunoprecipitation. Variables associated with assay conditions will dictate the optimal working dilution.

The aim of the present was to evaluate the effects of DNA methylation and histone acetylation on 2,3,7,8‑tetrachlorodibenzo‑p‑dioxin (TCDD)‑induced cleft palate in fetal mice. Pregnant mice (n=10) were randomly divided into two groups: i) TCDD group, mice were treated with 28 µg/kg

Autophagy activation improves the phenotype in mdx mice, a Duchenne muscular dystrophy (DMD) model, although the underlying mechanisms are obscure. We previously found that resveratrol, a strong inducer of autophagy, ameliorates the cardiac pathology of mdx mice. Autophagy could eliminate

Chromatin compaction mediates progenitor to post-mitotic cell transitions and modulates gene expression programs, yet the mechanisms are poorly defined. Snf2h and Snf2l are ATP-dependent chromatin remodelling proteins that assemble, reposition and space nucleosomes, and are robustly expressed in the brain.
