General description

FER (fer (fps/fes related) tyrosine kinase) gene encodes a non-transmembrane receptor tyrosine kinase belonging to the FPS/FES family. The protein has a molar mass of 94kDa and contains a short, basic C-terminal tail and a large N-terminal hydrophilic domain that is similar to the 92kDa protein encoded by the mammalian c-fps/fes proto-oncogene. It contains a single SH2 domain.

Application

Biochem/physiol Actions

FER (fer (fps/fes related) tyrosine kinase) contains a coiled coil sequence that interacts with a catenin-like substrate pp120 and mediates cell-cell signaling. It is localized to the cytoplasmic fraction and interacts with actin-binding protein cortactin to form a complex. It functions in the growth factor-dependent phosphorylation of cortactin and mediates signaling from the cell surface to the cytoskeleton. The Fer protein that is localized on microtubules phosphorylates platelet endothelial cell adhesion molecule-1, a part of intercellular junctions that mediates intracellular signaling cascades. It phosphorylates plakoglobin, a protein that links E-cadherin and α-catenin, and enhances the binding of plakoglobin to α-catenin, thereby increasing the transcriptional activity of the β-catenin-Tcf-4 complex that is commonly found in many epithelial tumor cells. It mediates phosphorylation of focal adhesion kinase in suspended hepatocytes leading to multiple membrane protrusions and regulation of cell morphology.
