General description

Early growth response protein 1 (UniProt P18146; also known as AT225, EGR-1, Nerve growth factor-induced protein A, NGFI-A, Transcription factor ETR103, Transcription factor Zif268, Zinc finger protein 225, Zinc finger protein Krox-24) is encoded by the EGR1 (also known as KROX24, ZNF225) gene (Gene ID 1958) in human. EGR-1 is the founding member of the early growth response (EGR) family of Cys2-His2-type zinc finger transcription factors that share a high homology in their DNA-binding region. EGR-1 is ubiquitously expressed in many cell types and binds to a consensus DNA motif (GCGGTGGGCG) in target genes, including c-Myc, cyclins D2 & G2, p19, TNF, IL-2, CD44, and ICAM-1. EGR1 is known as an immediate-early response gene due to the rapid kinetics of its induction by various signals, including growth factors, cytokines, and stress. Human EGR-1 is a 543-amino acid protein with three C2H2-type Zinc fingers (a.a. 338-362, 368-390, 396-418).

Application

Anti-EGR1 Antibody, clone 16A10.1 is an antibody against EGR1 for use in Western Blotting, Immunocytochemistry.
Immunocytochemistry Analysis: An 1:250 dilution of this antibody from a representative lot detected EGF-induced upregulation of nuclear EGR1 immunoreactivity in serum-starved HeLa and NIH/3T3 cells.

Quality

Evaluated by Western Blotting in HUVEC lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected VEGF-induced EGR1 expression in serum-starved HUVECs.

Target description

~80 kDa observed. 57.51 kDa calculated. The observed target band size is consistent with the reported apparent molecular weight (Krones-Herzig, A., et al. (2005). Cancer Res. 65(12):5133-5143; Josefsen, K., et al. (1999). Diabetologia. 42(2):195-203).
