QuEChERS样品制备方法
QuEChERS是一种分散固相萃取(dSPE)样品制备技术。QuEChERS是“快速、简便、经济、高效、耐用且安全”(Quick, Easy, Cheap, Effective, Rugged and Safe)的简称。此方法最初针对水果和蔬菜农残分析样品制备而开发,现可用于分析其他类型化学物质,包括PAH(多环芳烃)、PCB(多氯联苯)、PBDE(多溴联苯醚)和阻燃剂。这一概念于2002年推出,现已成为分析化学家协会(AOAC 2007.01)和欧洲标准化委员会(EN 15662:2008)官方方法指定内容。

QuEChERS样品萃取
溶剂萃取技术旨在最大限度提高基底基质中的分析物得率。此技术使用与水混溶的溶剂(通常为乙腈)和高浓度盐和/或缓冲剂,从样品基质中提取目标分析物。溶剂选择对于减少化合物共萃取非常重要。萃取步骤使用盐实现相分离。这类盐充当缓冲剂调节pH值,并从样品中析出水分。
QuEChERS纯化
样品纯化可减少后续分析过程中的干扰成分,毕竟干扰成分可能损坏分析仪器、扰乱分析物鉴定和定量结果。和传统的SPE管样品纯化方法不同,QuEChERS通过将大量SPE吸附剂与萃取提取物混合,促进纯化。纯化所用的吸附剂可去除与目标分析物共萃取的基质干扰成分。在进行GC-MS/MS或LC-MS/MS分析之前,可利用PSA、C18或GCB(石墨化炭黑)吸附剂去除糖、脂质、固醇、有机酸、蛋白质、类胡萝卜素、叶绿素和其他色素。可能需要专用Z-Sep吸附剂去除含颜料或脂肪的复杂基质。
