Specificity

Clone 1M19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human g-Tubulin. It targets an epitope within 16 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the C-terminal region of human g-Tubulin.

Application

Quality Control Testing

Evaluated by Western Blotting in U2OS cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Tubulin g-2 in U2OS cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Tubulin -2 in human colon tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Tubulin peptide with a KD of 3.4 x 10-6 in an affinity binding assay.

Anti-Tubulin g-2, clone 1M19 ZooMAb®, Cat. No. ZRB1123, is a recombinant Rabbit monoclonal antibody that specifically targets Tubulin g-2 and is tested for use in Affinity Binding Assay, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Tubulin g-2 chain (UniProt: Q9NRH3; also known as Gamma-2-tubulin, Gamma tubulin) is encoded by the TUBG2 gene (Gene ID: 27175) in human. Tubulin is the major constituent of microtubules. In humans five isoforms of tubulin have been described and are known as -tubulin, β-tubulin, -tubulin, -tubulin, and -tubulin. The g -chain is found at microtubule organizing centers such as the spindle poles or the centrosome. It is a component of the pericentriolar matrix that regulates / β chain minus-end nucleation, centrosome duplication, and spindle formation. It is ubiquitously expressed and is abundant in both the cytosol and the nuclear compartments of cells in all mammalian tissues. By binding to the β-tubulin subunit, it establishes the polarity of a microtubule leaving the -tubulin subunit exposed at the positive end. Although g-tubulin is reported to be only about 1% of the level of either or β-tubulin, it shares approximately 28-32% identity with a-tubulin and 32-36% identity with β-tubulin in various species. g -tubulin is reported to form two complexes known as the g-tubulin small complex and the larger g-tubulin ring complex and each complex contains a several g-tubulin complex proteins (GCPs). g -1 tubulin knockout mice survive only up to the morula/blastocyst stage; however, g-2 tubulin knockout mice remain viable and fertile. In human neuroblastoma cells, g-2 tubulin is shown to be a pro-survival factor and its levels are up-regulated during neural development and oxidative stress. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
