Specificity

Clone 3E14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects SMARCA2. It targets an epitope within 24 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal region of human SMARCA2.

Application

Quality Control Testing

Evaluated by Western Blotting in MCF-7 cell lysate.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected SMARCA2 in MCF-7 cell lysate.

Tested applications

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution of this antibody detected SMARCA2 in human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound SMARCA2 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Western Blotting Analysis): A 1:1,000 dilution from a representative lot detected SMARCA2 in lysate from THP-1 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SMARCA2 in U2OS cells.

Target description

Probable global transcription activator SNF2L2 (UniProt: P51531; also known as ATP-dependent helicase SMARCA2, BRG1-associated factor 190B, BAF190B, Protein brahma homolog, hBRM, SNF2-alpha, SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily A member 2) is encoded by the SMARCA2 (also known as BAF190B, BRM, SNF2A, SNF2L2) gene (Gene ID: 6595) in human. SMARCA2 is a member of the SNF2/RAD54 helicase family and belongs to the neural progenitors-specific chromatin remodeling complex and the neuron-specific chromatin remodeling complex (nBAF complex). It is involved in transcriptional activation and repression of select genes by chromatin remodeling. During differentiation from embryonic stem cells to neural progenitor cells, SMARCA4 expression gradually declines and SMARCA2 is upregulated and continues to be highly expressed in neurons. SMARCA2 also has a tumor suppressor function, and its expression is associated with good prognosis. Its high expression is associated with benign differentiated tumors. It is found to be silenced in a number of cancer cell lines and primary tumors. Heterozygous missense mutations in the SMARCA2 gene have been linked to Nicolaides-Baraitser syndrome (NCBRS) that is characterized by severe mental retardation. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
