Agarase Specificity

Agarose

Agarose is the principal neutral gelling component of agar extracted from algae. Agarose is a complex range of polysaccharide chains composed of alternating α-(1-3)-D-galactosyl-β-(1-4)-anhydro-L-galactosyl units.1

Synonym: agarose 3-glycanohydrolase

Agarase catalyzes the hydrolysis of 1,3-b-D-galactosidic linkages in agarose, giving the tetramer as the predominant product. 1

Alginate Lyase and Alginic Acid

Alginate Lyase and Alginic Acid

Composed of blocks of β-D-(1-4) mannuronic acid homopolymeric regions (MMMM…), α-L-(1-4)guluronic acid (GGGG…) homopolymeric regions, and alternating copolymer regions of β-D-(1-4) mannuronic acid - α-L-(1-4)-guluronic acid (GMGMGM…). Bacterial alginic acid can be acetlylated at the 2 or 3 positions on mannuronic acid.

Alginate Lyase

EC# 4.2.2.3
Synonyms: poly(β-D-manuronate) lyase

Alginate lyase cleaves at the β-(1-4)-D-mannuronic bonds residues to yield oligosaccharides with 4-deoxy-α-L-erythro-hex-4-enopyranuronosyl groups at their non-reducing termini.

Amylopectin & Glycogen

Polymers of α-(1-4)-D-glycopyranosyl units. Amylopectin has approximately 4% α-(1-6)-D-glycopyranosyl branching. Glycogen tends to have a higher frequency of branches of shorter length than amylopectin.

β-Amylase

Synonyms: 1,4-α-D-glucan maltohydrolase

β-Amylase catalyzes the exo-hydrolysis of 1,4-α-D-glucosidic linkages in polysaccharides resulting in the successive liberation of maltose units from the non-reducing ends of the chains.

Amyloglucosidase

EC# 3.2.1.3
Synonyms: glucan 1,4-α-glucosidase, glucoamylase, γ-amylase

Amyloglucosidase catalyzes the hydrolysis of terminal 1,4-linked α-D-glucose residues successively from the non-reducing ends of maltooligo- and polysacharides with release of β-D-glucose. Most forms of the enzyme can rapidly hydrolyze 1,6-α-D-glucosidic bonds when the next bond in the sequence is 1,4- and some preparations of this enzyme hydrolyze 1,6- and 1,3-α-D-glucosidic bonds in other polysaccharides.

α-Glucosidase

EC# 3.2.1.20
Synonyms: maltase

α-Glucosidase catalyzes the hydrolysis of terminal 1,4-linked α-D-glucose residues successively from the non-reducing ends of maltooligo- and to a lesser extent polysacharides with release of β-D-glucose. Most forms of the enzyme can slowly hydrolyze 1,6-α-D-glucosidic bonds.

Repeating linear polymer of two β-(1-4)-D-glycopyranosyl and one β-(1-3)-D-glycopyranosyl unit.

Polymer of β-(1-4)-D-glycopyranosyl units occurring as predominantly as cellotriose and cellotetraose separated by single β-(1-3)-D-glycopyranosyl units. Cross-linking can occur within the consecutive cellotriose regions.
