Specificity
Clone 1B3 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects SMCR8. It targets an epitope within 15 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal half of human Guanine nucleotide exchange protein SMCR8.
Application
Quality Control Testing
Evaluated by Western Blotting with His-ABP-tagged recombinant fragment of Human SMCR8.
Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected His-ABP-tagged recombinant fragment of Human SMCR8.
Tested Applications
Enzyme Immunoassay (ELISA) Analysis: A serial of dilutions from a representative lot detected His-ABP-tagged recombinant fragment of Human SMCR8.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SMCR8 in Caco-2 cells.
Affinity Binding Assay: A representative lot of this antibody bound SMCR8 peptide with a KD of 2.0 x 10-6 in an affinity binding assay.

Target description
Guanine nucleotide exchange protein SMCR8 (UniProt: Q8TEV9; also known as Smith-Magenis syndrome chromosomal region candidate gene 8 protein) is encoded by the SMCR8 gene (Gene ID: 140775) in human. Long isoform of human C9orf72 protein is known to form a complex with SMCR8 and WDR41 (WD Repeat domain 41) proteins. This complex displays guanine nucleotide exchange factor (GEF) activity and regulates autophagy. The N-terminal sequence (aa 1-349) of SMCR8 is required for its association with C9orf72. In the complex, C9orf72 and SMCR8 are suggested to constitute the catalytic subunits that promote the exchange of GDP to GTP, converting inactive GDP-bound Rab8A and Rab39B into their active GTP-bound form, thereby promoting autophagosome maturation. The C9orf72-SMCR8 complex is also reported to act as a negative regulator of autophagy initiation by interacting with the ULK1/ATG1 kinase complex and inhibiting its protein kinase activity. In addition to its activity in the cytoplasm within the C9orf72-SMCR8 complex, SMCR8 is also shown to localize in the nucleus, where it associates with chromatin and negatively regulates expression of suppresses ULK1 and WIPI2 genes. It is also reported to act as a regulator of mTORC1 signaling by promoting phosphorylation of mTORC1 substrates. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
