General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone 1C7 is a mouse monoclonal antibody that detects LIF-R (CD118). It targets an epitope within the extracellular domain.

Immunogen

Recombinant protein fragment corresponding to amino acids 2-538 of human LIF-R, human IgG Fc fusion protein.

Evaluated by Flow Cytometry in CHO cells stably transfected with D1IgD2-mycDAF.

Flow Cytometry Analysis: Staining of one million Staining of one million CHO cells stably transfected with D1IgD2-mycDAF was performed using 5 µL of a 1:1 mixture of Cat. No. CWA-1171, Anti-Human LIF-R (1C7) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye.

Tested Applications

No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Leukemia inhibitory factor receptor (UniProt: P42702; also known as LIF receptor, LIF-R, CD118) is encoded by the Lifr gene (Gene ID: 3977) in human. Leukemia inhibitory factor (LIF) mediates its biological actions via binding to its high affinity receptor made of the low-affinity LIF receptor subunit gp190 (LIF-R) and the gp130 subunit. LIF-R, a member of the type I cytokine receptor family, is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-44), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 45-833), a transmembrane domain (aa 834-858), and a cytoplasmic domain (aa 859-1097). It also contains six fibronectin type III domains. It contains a WSXWS motif (aa 519-523) that is shown to be essential for proper protein folding and cell-surface receptor binding. Its box 1 motif (aa 869-877) is required for JAK interaction and activation. A secreted form (isoform 2) has also been described that inhibits the biological activity of LIF by blocking its binding to receptors on target cells. LIF-R forms a heterodimer with interleukin 6 cytokine family signal transducer (IL6ST) and this heterodimer interacts with the complex formed by ciliary neurotrophic factor (CNTF) and CNTF receptor (CNTFR). Over-expression of LIF and LIF-R has been reported in many solid cancers. LIF/LIF-R activate oncogenic signaling pathways, including JAK/STAT3, MAPK, Akt, and mTOR. This signaling also plays a role in modulating multiple immune cell types present in tumor microenvironment. This ColorWheel® Dye-Ready monoclonal antibody was developed using our Cat. No. MABD150; Anti-LIF Receptor, clone 1C7. (Ref.: Viswanadhapalli, S., et al (2022). Genes Dis. 9(4); 973-980; Hisaka, T., et al. (2004). Comp. Hepatol.3(1); 10; Taupin, J-L., et al. (2001). J. Biol. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 µL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 µL PBS. Protect from light.
