General description

Pyruvate kinase (PK) has four isozymes such as PKL, PKR, PKM1, and PKM2. PKM1/2 is located in the distal convoluted tubule of the kidney, embryo, lungs, proliferative tissues, retina, adipose tissues, and islets. The PKM1 gene is located on the human chromosome at 15q23.

Application

PKM1active human has been used in a cell-based kinase assay to study the ability ofa compound to inhibit the kinase activity of PKM1. It has also been used inoxidative stress experiments on PKM2.

Biochem/physiol Actions

Pyruvate kinase (PK) catalyzes the dephosphorylation of phosphoenolpyruvate(PEP) into pyruvate. It is also involved in glycolysis by releasing ATP.Allosteric effects and intracellular signal transduction regulate the enzymaticactivity of PKM1. PKM1 is involved in several biological processes such asmetabolic reprogramming, and innate and acquired immune response. Itparticipates in protein interactions, protein synthesis, glycolysis, andintranuclear signal transduction.
