Specificity

Clone 1D12 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Gephyrin. It targets an epitope within 25 amino acids from the C-terminal half of the protein.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal half of human Gephyrin.

Application

Quality Control Testing

Evaluated by Western Blotting in Neuro-2a cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Gephyrin in Neuro-2a cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000-10,000 dilution from a representative lot detected Gephyrin in rat brain lysates and Myc/DDK-tagged recombinant human Gephyrin.

Enzyme Immunoassay (ELISA) Analysis: Serial dilutions from a representative lot detected Myc/DDK-tagged recombinant human Gephyrin.

Affinity Binding Assay Analysis: The binding affinity of this antibody to Gephyrin peptide was at least ten thousand-fold (10,000X) higher than that obtained with the non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Gephyrin (UniProt: Q9NQX3; also known as) is encoded by the GPHN (also known as) gene (Gene ID: 10243) in human. Gephyrin is a multifunctional enzyme and a multi-domain protein consisting of an N-terminal G-domain (Molybdopterin adenylyltransferase, EC:2.7.7.75), which interacts with receptors, a central linker region, and a C-terminal E-domain (Molybdopterin molybdenumtransferase, EC:2.10.1.1), which binds to microtubules. Gephyrin plays a critical role in the clustering and anchoring of inhibitory neurotransmitter receptors at the postsynaptic membrane of neurons. Its primary function is to cluster and localize glycine and gamma-aminobutyric acid (GABA) receptors at inhibitory synapses in the central nervous system. It interacts with these receptors through its PDZ domain, ensuring their proper positioning and functional integrity. Gephyrin is predominantly expressed in the brain and spinal cord, particularly in regions associated with inhibitory neurotransmission, such as the hippocampus and cerebellum. Its expression is crucial during brain development, contributing to synapse formation and maturation. Gephyrin can associate with microtubules in a subset of neuronal processes, potentially impacting neuronal morphology and intracellular transport. Mutations in the gene encoding gephyrin (GPHN) have been linked to certain types of neurodevelopmental disorders, including hyperekplexia and autism spectrum disorders. Hyperekplexia, also known as startle disease, is characterized by an exaggerated startle response, muscle stiffness, and an increased risk of sudden infant death syndrome (SIDS). These mutations disrupt gephyrin′s interaction with receptors, impairing inhibitory neurotransmission. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
