General description

Monoclonal Anti-CHD4 (mouse IgG2a isotype) is derived from the hybridoma 3F2/4 produced by the fusion of mouse myeloma cells and splenocytes from CD-1 mice immunized with a synthetic peptide derived from human chromodomain helicase DNA binding protein 4 (CHD4) conjugated to keyhole limpet hemocyanine (KLH). CHD4 is one of the subunits of Mi-2, an ATP-dependent helicase. It belongs to the sucrose non-fermentable (SNF2)/Rad54 helicase family of proteins. The CHD4 gene is mapped to human chromosome 12p13.31 and is highly conserved among animals and plants.

Biochem/physiol Actions

Chromodomain helicase DNA binding protein 4 (CHD4) interacts with metastasis-associated protein (MTA1/2), histone deacetylase 1 (HDAC1) and RbAp46 in the nucleosome remodelling and deacetylation (NuRD) complex. Interestingly, antibodies against CHD4 have been found to be involved in the onset of dermatomyositis (DM), an autoimmune disease which is a form of idiopathic inflammatory myopathy (IIM). Variation in the gene due to mutation is observed in endometrial cancer. CHD4 regulates homologous recombination and thus maintains genome stability.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
