General description

Autophagy-specific gene 8a and 8b (UniProt Q9W2S2 and Q9VEG5, respectively) are encoded by the drosophila Atg8a (ORF Dmel_CG32672; Gene ID 32001) and Atg8b (ORF Dmel_CG12334; Gene ID 42132) gene, respectively. Autophagy is a fundamental an adaptive response to cellular stress caused by, for example, starvation and viral infection. Autophagy is a process conserved from yeast to human and involves the formation of double-membrane vesicles for the purpose of recycling nutrients by means of degrading the engulfed content. In resting cells, the majority of Atg8 is in an unlipidated form called Atg8-I. During autophagy, Atg8-I is conjugated with a lipid moiety, and the lipidated form (Atg8-II) is recruited to autophagosomal membranes. An elevated Atg8-II level therefore serves as an indicator of autophagy activation. LC3 is the mammalian ortholog of drosophila Atg8.

Reacts with the unlipidated (Atg8-I) and lipidated (Atg8-II) forms of drosophila Atg8a and Atg8b (Shelly, S., et al. (2009). Immunity. 30(4):588-598).

This Anti-ATG8 Antibody is validated for use in Western Blotting for the detection of ATG8.
Western Blotting Analysis: A representative lot detected the induction of the lipidated Atg8-II band in wild-type flies upon Rift Valley fever virus (RVFV) infection, while an enhanced Atg8-II induction was observed in Toll-1-overexpressing flies and no Atg8-II induction by RVFV infection was seen among Toll-7 mutant flies (Moy, R.H., et al. (2014). Immunity. 40(1):51-65).
Western Blotting Analysis: A representative lot detected sdRNA-targeted downregulation of Atg8a and Atg8b in Drosophila S2 cells (Shelly, S., et al. (2009). Immunity. 30(4):588-598).
Western Blotting Analysis: A representative lot detected the 16 kDa Atg8-I band (unlipidated) as well as the serum starvation- and vesicular stomatitis virus (VSV) infection-induced 14 kDa Atg8-II band (lipidated) in Drosophila S2 cells (Shelly, S., et al. (2009). Immunity. 30(4):588-598).

Quality

Evaluated by Western Blotting in DL1 cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected ATG8 in 10 µg of DL1 cell lysate.

Target description

~16/14 kDa observed. The upper band of the doublet represents the unlipidated Atg8-I form, while the lower band of the doublet represents the lipidated Atg8-II form that is upregulated upon autophagy induction.
