General description

Zolpidem Phenyl-4-carboxylic acid is the major urinary metabolite of zolpidem, accounting for 51% of an administered dose. Literature references indicate the metabolite can be found in urine for 2-3 days following ingestion of a single therapeutic dose of zolpidem. Zolpidem, marketed as Ambien® or Stilnox®, is a nonbenzodiazepine used for the short-term treatment of insomnia.

Application

  • Pharmacokinetics in forensic contexts: A study demonstrated the simultaneous analysis of Zolpidem and its metabolites, including Zolpidem phenyl-4-carboxylic acid, in postmortem urine, which is critical for forensic toxicology (Yamaguchi et al., 2023).

  • Detection in hair samples: Research developed a sensitive LC-MS/MS method for detecting picogram levels of Zolpidem and its primary metabolites, such as Zolpidem phenyl-4-carboxylic acid, in hair, offering valuable insights for forensic and clinical investigations (Kim et al., 2021).

  • Advanced analytical techniques: The determination of Zolpidem phenyl-4-carboxylic acid and Zolpidem 6-carboxylic acid in hair was achieved using gas chromatography-electron ionization-tandem mass spectrometry, enhancing the capabilities of toxicological analyses (Kwon et al., 2021).

  • Pharmacokinetic profiling: A study on the pharmacokinetics of Zolpidem and Zolpidem phenyl-4-carboxylic acid in oral fluid post administration provided crucial data for understanding drug behavior in therapeutic monitoring (Feng et al., 2019).

  • Forensic case study: An examination of a fatal zolpidem poisoning case highlighted the postmortem distribution and redistribution of Zolpidem and its predominant metabolite, Zolpidem phenyl-4-carboxylic acid, in body fluids and tissues, offering insights for forensic pathology (Hasegawa et al., 2018).

Zolpidem is a non-benzodiazepine hypnotic that has been implicated in both drug-facilitated sexual assault and drink spiking. Detection of the drug in urine is extremely difficult because of its extensive metabolism. A method is presented for the detection and quantitation

An rapid method for the siµLtaneous determination of the Z-drugs or sleep aids: zopiclone, zolpidem, and zaleplon is presented here. The need for greater analytical capacity and throughput for the analysis of sleep aid medicines (Z-drugs) in forensic toxicology laboratories can be met by the use of fast Ascentis Express 2.0 micron Fused Core UHPLC Columns.
