Lentiviral Transduction

In this exercise we will be demonstrating lentiviral transduction on two different cell lines. In the first example the human neuroblastoma cell line SK-N-SH will be transduced with MISSION TurboGFP Control particles. A range of MOI’s will be tested and an increase in GFP positive cells will be observed with an increase in MOI.

The second cell line we will be using is the Chinese hamster ovary cell line CHO-K1 that has previously been engineered to exhibit stable TurboGFP expression. In this exercise we will transduce these cells with the MISSION TurboGFP shRNA with the goal of repressing GFP expression. A range of MOI’s will be tested and a decrease in GFP expression will be observed with an increase in MOI.

We will be using a 24-well culture plate seeded with 15,000 SK-N-SH cells per well as shown on the top row and 15,000 CHO-K1 GFP cells per well on the bottom row of the diagram.
