Description générale

Beta-nerve growth factor (UniProt: P01139; also known as Beta-NGF) is encoded by the Ngf (also known as Ngfb) gene (Gene ID: 18049) in murine species. β-NGF, an endogenous neurotrophic factor, is a homodimeric glycoprotein that is synthesized with a signal peptide (aa 1-18) and a propeptide (aa 19-121) that are subsequently cleaved off to generate the mature form (aa 122-241). It is highly expressed in male submaxillary gland and its levels are much lower in female submaxillary gland. Its expression is reported to oscillate in a circadian manner in the suprachiasmatic nucleus (SCN) of the brain. NGF is important for the development and maintenance of the sympathetic and sensory nervous systems. It serves as an extracellular ligand for the NTRK1 and NGF receptors to activate cellular signaling cascades to regulate neuronal proliferation, differentiation, and survival. However, it does not permeate the blood-brain barrier. Its precursor form (ProNGF) is reported to function as a ligand for the heterodimeric receptor formed by Sortilin Related VPS10 Domain Containing Receptor 2 (SORCS2) and NGF receptor that activates signaling cascade leading to inactivation of RAC1 and/or RAC2. In contrast to the mature form, ProNGF is known to promote neuronal apoptosis. N-glycosylation of ProNGF is reported to be essential for its processing, secretion, and construction of the tertiary fold of the β-NGF homodimer. Clone D11 is a potent antagonist that can effectively antagonize rodent and human NGF in a variety of in vitro and in vivo systems. (Ref.: Covaceuszach, S., et al. (2012). Spécificité

Clone D11 is a rat monoclonal antibody that detects Nerve growth factor (NGF). It targets an epitope within 23 amino acids from the N-terminal half of the mature NGF.

Stockage et stabilité

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
