Specificity

Clone 1D6 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Collagen VI 3. It targets an epitope within 25 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal half of human Collagen VI 3.

Application

Quality Control Testing

Evaluated by Western Blotting in HT-1080 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Collagen VI 3 in HT-1080 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Collagen VI 3 in Human skeletal muscle, and Rat testis lysates.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected Collagen VI 3 in Human breast cancer tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Collagen VI 3 in Human breast cancer tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Collagen VI 3 peptide with a KD of 3.0 x 10-7 in an affinity binding assay.

Target description

Collagen alpha-3(VI) chain (UniProt: P12111; also known as Collagen alpha-3) is encoded by the COL6A3 gene (Gene ID: 1293) in human. The collagen super family includes over 20 different types of collagens with at least 38 distinct polypeptide chains. Of these types I, II, and III are the most abundant and are involved in the formation of fibrils. Collagen -3(VI) is a secreted protein that is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to produce the mature form that is highly glycosylated. It is a breaded filament type of collagen that is found in most connective tissues. Collagen VI is reported to first assemble into a triple helix monomer by the - 1 (VI), 2 (VI) and 3 (VI) chains in a 1:1:1 stoichiometric ratio. The -3 chain is shown to be much larger than the 1 and 2 chains. The triple helix monomers are staggered into anti-parallel dimers bonded by disulfide bonds, and the anti-parallel dimers are also aligned through disulfide bonds to form tetramer. These tetramers are secreted to the extracellular matrix and form beaded microfibrils. Mutations in COL6A3 gene are associated with Bethlem myopathy, a rare autosomal dominant proximal myopathy with onset in childhood onset. Some mutations also cause Ullrich congenital muscular dystrophy, an autosomal recessive congenital myopathy that is more severe than Bethlem myopathy. Five isoforms of Collagen -3(VI) have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

30 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
