General description

Chromozym® PK is a non-radioactive amidolytic chromogenic substrate mostly used in kinetic analysis of plasma kallikrein.

Application

Use Chromozym® PK as a substrate for the determination of serine proteases, specifically plasma kallikrein in citrated plasma.

Sequence

Benzoyl-prolyl-phenalanyl-arginine-4-nitranilide-acetate

Principle

Chromozym PK is cleaved by plasma kallikrein into a residual peptide and free 4-nitraniline, which is measured at 405 nm. The absorbance difference per minute is used for the determination of the kallikrein activity in U/ml.

Preparation Note

Activator: Mg2+ (Km = 3.0 mM, pH 7.4, 25 °C); other divalent metal ions (Mn2+, Fe2+, Zn2+)
Working concentration: Approximately 0.5 mM
Storage conditions (working solution): 2 to 8 °C
An aqueous solution of Chromozym PK (1.3 mM) is stable for at least 4 weeks if stored at 2 to 8 °C.
