Gene Information

human ... SIRT1(23411)
mouse ... Sirt1(93759)

General description

Sirtuins are a family of closely related enzymes named after the yeast gene Sir2 (silent mating type information regulation 2). Members of the Sirtuin family have been found in nearly all organisms studied. Sirtuins are hypothesized to play a key role in an organism′s response to stresses (such as heat or starvation) and to be responsible for the lifespan-extending effects of calorie restriction. Sirtuins act by removing acetyl groups from proteins in the presence of NAD+; they are thus classified as "NAD+-dependent deacetylases". Sirtuins have been implicated in the regulation of aging, transcription, apoptosis and stress resistance. Sirt1 (also known as Sirtuin 1, Sir2L1, and Sir2a), the mammalian homolog of yeast Sir2, has been shown to deacetylate and thereby deactivate the p53 protein to regulate cellular processes.

Specificity

This antibody recognizes Sirt1.

Immunogen

Epitope: a.a. 1-131
GST-tagged fusion protein corresponding to amino acids 1-131 of mouse Sir2α.

Application

Immunoprecipitation:
5 µL of a previous lot of this antibody has been shown to immunoprecipitate Sirt1 from 1 mg of a 3T3/A31 RIPA cell extract.

Immunocytochemistry:
This antibody has been reported to detect Sirt1 in 3T3 cells fixed with paraformaldehyde.

Chromatin Immunoprecipitation:

Use Anti-Sirt1(Sir2) Antibody (Rabbit Polyclonal Antibody) is published (more than 30 citations) and validated in ICC, IP, WB to detect Sirt1(Sir2) also known as Silencing information regulator 2-like, sirtuin 2.

Quality

Routinely evaluated by western blot on 3T3 nuclear extract.

Western Blot Analysis:
A 1:1000-1:5000 dilution of this lot detected Sirt1 in a 3T3 nuclear extract.

Target description

110 kDa; Uncharacterized bands 70 and 50 observed in some Lysates.

Linkage

Replaces: 05-707

Physical form

Immunoaffinity Chromatography
Purified rabbit IgG in buffer containing 0.1M Tris-Glycine, 0.15M NaCl, 0.05% sodium azide and 30% gylcerol

Storage and Stability

1 year at -20°C from date of shipment.
Handling Recommendations: Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C.
Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.

Analysis Note

Control
Jurkat cell lysate, human colon carcinoma or human lung squamous carcinoma.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
