General description

Adeno-associated virus (AAV) is a small (25-nm), non-enveloped virus that packages a linear single-stranded DNA genome. It belongs to the family Parvoviridae and productive infection by AAV occurs only in the presence of a helper virus, which could either be adenovirus or herpesvirus. After successful infection, two stages of AAV life cycle have been described - a lytic stage and a lysogenic stage. In the presence of helper virus, the lytic stage ensues and during this period, AAV undergoes productive infection with genome replication, viral gene expression, and virion production. The lysogenic stage is established in host cells in the absence of a helper virus and AAV integrates into the host genome at specific site, AAVS1 on human chromosome 19. This site is favored due to the presence of a Rep binding element. AAV genome consists of two open reading frames, Rep and Cap, flanked by two 145 base inverted terminal repeats (ITR). The ITR base pairs allow for synthesis of the complementary DNA strand. Rep and Cap are translated to produce multiple distinct proteins. The Rep gene encodes Rep78, Rep68, Rep52, Rep40 and Cap gene produces VP1, VP2, VP3 capsid proteins. The Rep78 and Rep68 proteins participate in DNA replication process via their interaction with Rep-binding element (RBE) and terminal resolution site sequences located within the ITR. The Rep52 and Rep40 proteins are involved in the generation and accumulation of single-stranded viral genomes from double-stranded replicative intermediates. Thus far eleven serotypes of AAV have been identified, which differ in their tropism or the type of cells they infect. AAV8 shown to be suitable for use hepatic cell transduction and can serve as a useful vector for studies involving the hepatic tissue. Clone HL2381 generated against the AAV8 capsid, binds to AAV8, but also displays cross-reactivity with AAV3B. It can also neutralize infection in cells. (Ref.: Russell, WC et al. (2009). J. Gen. Specificity

Clone HL2381 is a mouse monoclonal antibody that detects AAV8 capsid protein. Also displays cross-reactivity with AAV3B capsid protein.

Application

Anti-Adeno-associated virus 8, clone HL2381, Cat. No. MABF2305, is a mouse monoclonal antibody that detects Adeno-associated viruses serotype 8 and has been tested for use in Dot Blot, ELISA, and Neutralizing Applications.

Quality

Evaluated by Dot Blot with AAV8 and AAV9 virus like particles .

Dot Blot Analysis: Various dilutions of AAV8 and AAV9 virus like particles were probed with Adenovirus 8. This antibody detects AAV8, but not AAV9.
