Specificity

Clone 1C9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects X-ray repair cross-complementing protein 5 (XRCC5). It targets an epitope within 15 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human X-ray repair cross-complementing protein 5 (XRCC5/Ku80).

Application

Quality Control Testing

Evaluated by Western Blotting in U2OS cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected XRCC/Ku80 in U2OS cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected XRCC/Ku80 in A549 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected XRCC/Ku80 in human testis tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected XRCC/Ku80 in U2OS cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

X-ray repair cross-complementing protein 5 (UniProt: P13010; also known as 86 kDa subunit of Ku antigen, ATP-dependent DNA helicase 2 subunit 2, ATP-dependent DNA helicase II 80 kDa subunit, CTC box-binding factor 85 kDa subunit, CTC85, CTCBF, DNA repair protein XRCC5, Ku80, Ku86, Lupus Ku autoantigen protein p86, Nuclear factor IV, Thyroid-lupus autoantigen, TLAA, X-ray repair complementing defective repair in Chinese hamster cells 5) is encoded by the XRCC5 (also known as G22P2) gene (Gene ID: 7520) in human. XRCC5 is a single stranded DNA-dependent ATP-dependent helicase that plays a key role in DNA non-homologous end joining (NHEJ) by recruiting DNA-dependent protein kinase (DNA-PK) to DNA. Its Ku domain is localized in amino acids 251-460. It also contains an EEXXXDL motif (aa 720-728) that is which is required for the interaction with catalytic subunit PRKDC and its recruitment to sites of DNA damage. It exists as a heterodimer composed on XRCC5/XRCC6 (Ku80/Ku70). This dimer acts as regulatory subunit of the DNA-PK that increases the affinity of the catalytic subunit PRKDC to DNA by about 100-fold. This dimer is also suggested to act as a 5′-deoxyribose-5-phosphate lyase (5′-dRP lyase), by catalyzing the beta-elimination of the 5′ deoxyribose-5-phosphate at an abasic site near double-strand breaks. Here, XRCC5 probably acts as the catalytic subunit of 5′-dRP activity and allows to ′clean′ the termini of abasic sites. In association with N- -acetyltransferase 15 (NAA15), this dimer is reported to bind to the osteocalcin promoter and activates osteocalcin expression. In murine cells absence of either of the subunits leads to increased radiosensitivity, defective DNA double-strand break repair, and defective in V(D)J recombination. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

30 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
